Most desktop websites don't use JQuery Mobile, in fact not many do at all. If your intention is to have a device agnostic website with minimal development, use JQuery Mobile and it will look the same on your mobile device as it does on the desktop.

The jQuery Mobile documentation is a good example of a desktop site built using jQuery Mobile.

The "look and feel" is no different between desktop and mobile, so I'm not sure what the point of your inquiry is.

Most of us building mobile sites want them to look good on both phones and tablets, and if it works well on a tablet it will work well on a desktop as well.

If you want to differentiate further, that's up to you.
